目的观察碘、氟对大鼠子代脑发育形态学方面是否存在联合作用.方法将实验用Wistar成年大鼠雌16只,雄8只,按饮水中不同碘、氟或碘+氟浓度分为8组,即:碘、氟为0(对照组,采用重蒸水),碘0.5、1.0、2.0mg/L,氟50 mg/L,碘+氟0.5+50、1.0+50、2.0+50 mg/L.喂养90 d后相同浓度组雌、雄按2∶1合笼交配,交配后2l~28 d母鼠产仔,哺乳5 d后将各组仔鼠断头处死.取其脑组织进行大脑重量、光镜(观察大脑皮质神经细胞密度及未分化神经母细胞占细胞总数百分比)、电镜(观察神经元发育情况)等形态学指标检查.结果仔鼠大脑重量及光镜指标经2×4析因方差分析均表明各组之间差异有显著性(P<0.001).其中氟50 mg/L、碘2.0 mg/L及碘+氟2.0+50 mg/L组各项指标都提示仔鼠大脑发育状况次于其余各组.电镜检查发现氟50 mg/L及碘+氟2.0+50 ml/L组神经元发育及超微结构形态有轻微变性改变.结论长期饲以高氟及高碘饮水的大鼠均可影响其子代大脑发育,且高碘、高氟对仔鼠大脑发育的危害存在协同作用.
Objective: To observe whether there is a combined effect of iodine and fluorine on the morphological development of rat offspring. Methods: Sixteen female and eight male Wistar adult rats were divided into eight groups according to different iodine, fluorine or iodine + fluorine concentrations in drinking water, namely: 0 iodine and fluorine (control group, using redistilled water), 0.5, 10, 20 mg/L iodine, 50 mg/L fluorine, 0.5 + 50, 10 + 50, 20 + 50 mg/L iodine + fluorine. After 90 days of feeding, the males and females in the same concentration groups were mated in a 2:1 ratio. The females gave birth 21 to 28 days after mating. After 5 days of lactation, the offspring in each group were decapitated and killed. The brain tissues were obtained for examination of morphological indicators such as brain weight, light microscopy (observation of the density of cerebral cortical neurons and the percentage of undifferentiated neuroblasts in the total number of cells), and electron microscopy (observation of neuronal development). Results :The brain weight and light microscopic indexes of offspring rats showed significant differences among the groups by 2×4 factorial analysis of variance (P <0 0 0 1). The indexes of the fluorine 50mg/L, iodine 20mg/L and iodine + fluorine 20+ 50mg/L groups indicated that the brain development of offspring rats was inferior to that of the other groups. Electron microscopic examination revealed that there were slight degenerative changes in neuronal development and ultrastructural morphology in the fluorine 50mg/L and iodine + fluorine 20+ 50ml/L groups. Conclusion: Long-term feeding of rats with high fluorine and high iodine drinking water can affect the brain development of their offspring, and high iodine and high fluorine have a synergistic effect on the brain development of offspring rats.
