【摘要】:目的探讨高氟对人甲状腺细胞功能的影响。方法用人甲状腺细胞,采用原代培养方法,按氟(NaF)剂量不同分为:0(对照组)、40、80、160 mg/L组。染氟48 h后,采用噻唑蓝(MTT)法测定细胞存活率,化学发光法测定抗甲状腺过氧化物酶抗体(TPOA)水平和荧光免疫分析法测定血清总甲状腺激素(T_4)水平。结果甲状腺细胞染氟培养48 h后,细胞存活率组间比较差异有统计学意义(P0.05);40 mg/L组细胞存活率与对照组(100)%比较,差异无统计学意义(P0.05);80、160 mg/L组细胞存活率分别与对照组比较,差异有统计学意义(P0.05)。甲状腺细胞T_4水平随着染氟剂量的升高而明显降低,组间比较差异有统计学意义(P0.05)。甲状腺细胞TPOA表达水平也随着染氟剂量的升高而明显增加,组间比较差异有统计学意义(P0.05)。结论高氟可损伤甲状腺细胞,降低细胞存活率,提高TTPOA表达,降低T_4水平。

Abstract: Objective To investigate the fluoride on human thyroid cell function. Methods employing thyroid cells, using primary culture method, according fluoride (NaF) is divided into different doses: 0 (control group), 40,80,160 mg / L group. After 48 h exposed to fluoride, using MTT (MTT) cell viability was determined, chemiluminescence anti-thyroid peroxidase antibodies (TPOA) levels and fluorescence immunoassay method for measuring serum total thyroid hormone (T_4) levels. Results thyroid cells after 48 h culture exposed to fluoride, cell survival difference between groups was statistically significant (P0.05); 40 mg / L group, cell viability and the control group (100)%, the difference was not statistically significant ( P0.05); 80,160 mg / L of cell survival rates were compared with the control group, the difference was statistically significant (P0.05). T_4 levels of thyroid cells transfected with elevated fluoride doses significantly reduced, the difference between groups was statistically significant (P0.05). TPOA thyroid cells transfected with expression levels were also elevated fluoride doses significantly increased, the difference between groups was statistically significant (P0.05). Conclusion fluoride can damage the thyroid cells, reducing cell viability, improve TTPOA expression T_4 lower level.
